Inclusion of relevant environmental cues is critical for tuning the cellular microenvironment. Conjugation of desired biomolecules to the surfaces of or distributed throughout the microgel particles at known concentrations and distributions is available with x_LLS™, making it one of the only chemically defined 3D culture mediums on the market. For example, LLS™ coated with collagen I (COL1-LLS) promotes cellular adhesion and migration and is an excellent medium for immunotherapy studies. Internal fluorescent tags can be added to the particles in order to make them visible in fluorescence microscopy for imaging studies or even to be detected in flow cytometry. Sold in 7 ml aliquots.

COL1-LLS has been shown to be highly effective for promoting cellular adhesion and is available on demand. Also available is RhDCOL1-LLS, which is collagen coated LLS™ with internal fluorescence from covalently bound rhodamine. Custom combinations of surface coating and internal fluorescence are available upon request. All existing and new combinations are quality checked for size, modulus, and surface coverage. A data sheet is provided with each batch.
Staining, fixation, dosing with drugs, or administration of other soluble factors is all possible in LLS™ by either perfusion in a Darcy Plate™, diffusion in static samples, or with periodic mixing for pre or post-perfusion operations. The mesh size of the hydrogel particles is about 7 nm, making the particles permeable to most small molecules while excluding larger species like antibodies or viruses.
LLS™ is shipped in excess liquid (1X PBS), and should be equilibrated with the culture medium of choice before use by centrifuging the mixture, replacing supernatant with media, then mixing and allowing to sit for at least 2 hours. LLS™ should be stored with excess liquid at 4ºC and away from direct sunlight.
