The 48-Well Darcy Plate™ is a single-use, perfusion-enabled 3D culture plate which supports a variety of samples when used alongside Liquid-Like Solids (LLS™) including adherent and non-adherent cells, explants, organoids, and other microorganisms. No external pressure device is required to operate this microplate; fluid height differences between the feed well and sample well drive perfusion through the LLS™ volume over 6-8 hours.

Darcy perfusion devices support extended culture of a variety of samples through passive perfusion, or the flow of cell culture media through the permeable Liquid-Like Solids (LLS™) 3D culture medium. LLS™ mimics a capillary bed by providing interconnected fluid channels through the soft hydrogel particles. These channels facilitate perfusion while also creating spaces for cellular migration and growth. These properties make LLS™ ideal for a variety of applications, including both metabolically active high-density cell cultures and Aurita's In Vitro ImmunoTherapy Assays (iVITA™).
The 48-Well Darcy Microplate™ achieves passive perfusion via differences in fluid height between the feed and sample wells. An estimated 6-8 hours of perfusion time is achievable when effluent is removed from the sample well and the feed media is replenished to full. A convenient shelf is included in each sample well for pipette tips to index off of during aspiration.
In situ dosing of cells with perfusion is possible throughout an experiment by replacing the media in the feed reservoir with the media solution including the soluble factors to be administered to the cells and then continuing perfusion.
